THE INTRODUCTION OF GROWTH HORMONE IN MURASHIGE AND SKOOG MEDIA MODIFICATIONS TO THE GROWTH OF POTATO PLANLET (SolanumtuberosumL.) USING THE VARIETY OF ATLANTIC, GRANOLA ANDMEDIANSEllenSimatupang, ReubenSuwitono,JuniartiSahat,DoliSitumeang, JoshuaTobing
Rice consumption of the people of Indonesia is 100,75 kg per person per year, the highest compare with Malaysia, Vietnam, and Thailand.Thepopulation of Indonesia is 238,5 million people in 2010, and twenty five years later will be 305,6 millionpeoplein 2035. In this case, there is a need for the diversification of staple food, not only rice, but also potato as the carbohydrate source of food. For thethedevelopment of potato, the implementation of tissue culture is promising because it can give us the high quality plant in mass production, and it helps eliminate plant diseases particularly virus.
InIndonesia, potato can be the substitute for carbohydrate sources after rice which is the staple food.Potatois one of the vegetables that has significant nutritional value despite of the fact that it is one of the carbohydrate source of food for many people.
The goal of this research is to know the influence of Hormones NAA, BAP, and GA3, and non hormone in the growth of roots and height in potatoplanlet.
Based upon the information that has been given, there are some problem identification such as :Is there any influence of the hormones and non hormones to the amount of roots in potatoplanlet?Is there any difference between some potato variety namely Granola, Atlantic, and Median in terms of the amount of roots inpotateplanlet?Is there any influence of hormones, non hormones, and the variety of potatoes in the amount of roots in potatoplanlet?
Basedupon the information that has been given, there are some problem identification such as :Isthere any influence of hormones and non hormones in the growth of height in potatoplanlet?Isthere any different between the three variety of potato in the growth of height in potatoplanlet?Is there any influence of hormones, non hormones, and the variety of potatoes in the growth of height in potatoplanlet?
Thematerials in this research are the three variety of potato namely : Granola,Atlantic,and Medians ; hormone BAP, NAA, GA3, MS Media,aquades,bacto-jell ,KOH, buffer, HCL, alcohol 96%.Sterilizationis very important in the tissue culture procedures. The room has to be clean all the time. Mapping the floor withdisinfectant,and clean the walls with alcohol 96%. The air in the room has to be very clean, free from dust.Autoclaveis the tool used to sterilize all theequipmentto be used such as bottles,clamp,scissors, scalpel, petri-dish, and pipet. Laminar Air Flow Cabinet also to be sterilized.
The making ofMedia, mediaused is MS orMurashigeandScoog2,2 gr with 4 gr ofBactoAgar.Theingredients was mixed in anerlenmeyerand heated until thephis 7. Then mixed with hormones NAA 1 mg, BAP 3 mg, and GA3 0,1 mg, then heated for 30 minutes until boiled.Preparationof potato explant with three variety in three different bottles. One bottle content 3 explantsObservationfor 3 weeks for data analysis.
Itwas observed that there were influence of hormones NAA, BAP, and GA3 to the number of roots in potatoplanlets.There is a difference of the amount of roots among the three variety of potatoes namelyAtlantic,Granola, and Medians.The interaction of hormones, non hormones, andthe three variety ofpotatoesgive influence to the number of roots of potatoesThere is an influence of hormones, NAA, BAP, and GA3, non hormones or control grp, to the height of potatoplanletsThere is a difference of height of the potatoplanletsamong the three variety of potato namelyAtlantic,Granola, and MediansThe interaction of hormones, non hormones, and the three variety of potatoes give influence to the height of the potatoplanlets.
